Characterization of a gonad-specific transforming growth factor-β superfamily member differentially expressed during the reproductive cycle of the oyster Crassostrea gigas

چکیده
Through differential screening between oyster families selected for high and low summer survival, we have characterized a new transforming growth factor-β (TGF-β) superfamily member. This novel factor, named oyster-gonadal-TGFβ-like (og-TGFβ-like), is synthesized as a 307 amino acid precursor and displays 6 of the 7 characteristic cysteine residues of the C-terminal, mature peptide. Sequence comparison revealed that og-TGFβ-like has a low percentage of identity with other known TGF-β superfamily members, suggesting that og-TGFβ-like is a derived member of this large superfamily. Real-time PCR (RT-PCR) analysis in different oyster tissues showed that og-TGFβ-like is specifically expressed in both male and female gonads, at distinct levels according to the reproductive stage. OgTGFβ-like relative expression was the lowest at the initiation of the reproductive cycle and increased as maturation proceeded to achieve a maximal level in fully mature female and male oysters. In situ hybridisation demonstrated that expression was exclusively detected in the somatic cells surrounding oocytes and spermatocytes. The role of this newly-characterized TGFβ member in the reproduction of cupped oyster is discussed in regard to the specificity and the localization of its expression, which singularly contrasts with the pleiotropic roles in a variety of physiological processes commonly ascribed to most TGF-β family members identified so far.

متن کاملImpact of food availability on energy storage and defense related hemocyte parameters of the Pacific oyster Crassostrea gigas during an experimental reproductive cycle
The aim of this study was to test the effect of food quantity on energy storage and defense capacities of oysters during a reproductive cycle. One-year-old Crassostrea gigas oysters were fed two different dietary rations (4% and 12% of oyster dry weight in algal dry weight per day) in controlled experimental conditions over an annual cycle. Oyster dry weights, carbohydrate and lipid contents, e...
متن کاملGametogenesis in the Pacific Oyster Crassostrea gigas: A Microarrays-Based Analysis Identifies Sex and Stage Specific Genes
BACKGROUND The Pacific oyster Crassostrea gigas (Mollusca, Lophotrochozoa) is an alternative and irregular protandrous hermaphrodite: most individuals mature first as males and then change sex several times. Little is known about genetic and phenotypic basis of sex differentiation in oysters, and little more about the molecular pathways regulating reproduction. We have recently developed and va...
متن کاملA Rhodopsin-Like Gene May Be Associated With the Light-Sensitivity of Adult Pacific Oyster Crassostrea gigas
Light-sensitivity is important for mollusc survival, as it plays a vital role in reproduction and predator avoidance. Light-sensitivity has been demonstrated in the adult Pacific oyster Crassostrea gigas, but the genes associated with light-sensitivity remain unclear. In the present study, we designed experiments to identify the genes associated with light-sensitivity in adult oysters. First, w...
متن کاملTemperature and photoperiod drive Crassostrea gigas reproductive internal clock
This study examined the gametogenic cycle of Crassostrea gigas in controlled conditions over one year, with a focus on the initiation of gametogenesis. This work analysed also the role of temperature and photoperiod in the regulation of oyster reproduction. Broodstock were maintained in natural (NC), accelerated (AC) and perpetual winter (WC) conditions of temperature and photoperiod, with feed...
